I think os.path.join(os.environ["SystemRoot"], "system32") should be fairly reliable. If people are worried that the directory might not be system32, then HKEY_LOCAL_MACHINE\SYSTEM\CurrentControlSet\Control\Windows\SystemDirectory gives you the value (although I could not find out how to expand REG_EXPAND_SZ keys with _winreg).
